Walton Jr, who injured his left hamstring against the Brisbane Bullets early in the first quarter, took no further part in the contest after consultation with the Phoenix medical staff.
Scans revealed the extent of the injury on Monday morning.
Phoenix General Manager of Basketball Operations Simon Mitchell said while the injury is devastating for the guard and the team, the Phoenix remain hopeful to see him again this campaign.
"Derrick has worked incredibly hard to recover from his earlier injury and played a pivotal role in the resurgence of the team in the past few months, so it's a real blow to have him go down again and we will support him in every way possible in his recovery," Mitchell said.
"With that said, we are hopeful to see Derrick on-court again for us this season as we eye off a post-season appearance, though his recovery and how his hamstring responds to treatment will dictate the timeline of return."
Walton Jr has been moved to the inactive list, with development player Luke Rosendale elevated to the main roster in his place.
The Phoenix take on Cairns tonight, ahead of three home games at the State Basketball Centre.
